Phospholipase D (PLD) catalyzes the hydrolysis of the phosphodiester bond of glycerophospholipids to generate phosphatidic acid and choline. It is ubiquitously expressed and is implicated in immune function and cancer inhibition.[1] Abnormalities in PLD expression have been associated with human cancers such as breast, renal, gastric and colorectal cancer.[2]
Phospholipase D (PLD) Activity Assay Kit has been used to measure PLD activity in the liver and lysosomal fractions of the mice liver.[3]

In this assay, PLD hydrolyzes phosphatidylcholine to choline, which is determined using choline oxidase resulting in a colorimetric (570 nm)/fluorometric (λex = 530/λem = 585 nm) product, proportional to the PLD activity in the sample. Unit definition: one unit of PLD catalyzes the formation of 1 mmole choline per minute under the assay conditions (pH 7.4).
